Which mod gets more power?

Headers or E85? Which mod gets better performance on an LT1 engine?

Both will require a tune to get the most out of them.... normally will void your powertrain warranty (depending on your local GM dealer). Are you OK with that? Personally, I got more HP/Torque from the header installation. Like the previous person stated, why not both? Flex Fuel kits are relatively inexpensive and the LT1 motor loves E.

E85 is definitely cheaper if one or the other. If going headers already though extra 300$ that the flex fuel kit cost is a no brainer.

Technically,

The average gains are about the same. But as others have said either requires a tune, but E85 tends to do better with a tune by not by much, and with both you are looking at about 40 hp maybe 50 hp give or take maybe a little more. Also depends on how good the tuner is as well.

If i could chose only one... id go E85... but because it requires a tune, id save up to do as many other mods as possible to take advantage of that investment. Consider a CAI, and Headers at the same time... for maximum bang for your buck.
